The reference occurred only in the descriptive part of the script, and was not said by any character.
Though the Jedi were not associated with monarchy in the second draft, they were seen as having families and lineages. This seems to be a carry-over from George Lucas's earlier versions, more closely derived from Akira Kurosawa's Hidden Fortress. The Jedi of the original story synopsis, rough draft and first draft were highly trained warriors with no obvious mystical abilities. They were, in fact, based on the samurai of pre-modern Japan, a social caste; being a samurai was based on heredity. Thus the implication of a Jedi prince is a "Jedi heir".
